چکیده

The rate of diffusion of a protein in solution from a source in agar gel is, within limits, proportional to its original concentration (Ouchterlony, 1949). When plasminogen in biological fluids is allowed to diffuse from a source in agar/fibrinogen gel for a given time, the distance diffused can be ascertained by activation of the plasminogen to plasmin by streptokinase after diffusion has taken place. The plasmin produces lysis of the fibrinogen in the gel which can easily be seen, thus the distance of the zone of lysis from the plasminogen source can be measured and the rate of diffusion calculated. This in theory would measure plasminogen concentration instead of plasmin activity. The effect of inhibitors of plasminogen activation and plasmin activity should be greatly reduced or eliminated if only the distance the plasminogen diffused and not the amount of lysis could be measured. Several methods can be devised on these principles. Unfortunately, however, it is extremely difficult to

Preparation of Plasminogen by Affinity Chromatography

Background: Plasminogen is one of the compounds derived from human plasma. Activation of plasminogen produces plasmin. Plasmin is able to lyse fibrinogen, fibrin, and some other human plasma proteins. The aim of the present work was to study the separation of human plasminogen by affinity chromatography using gel lysine Sepharose. Materials and Methods: Normal human plasma was used as the...

A study of proteases and protease-inhibitor complexes in biological fluids

We have (a) screened a variety of cell lines and body fluids for plasminogen activators and (b) studied the activity of proteases bound to alpha2- macroglobulin after exposing the complexes to partial degradation and/or denaturing procedures to unmask proteolytic activity. The respective results show (a) that the plasminogen activators in urine and cell culture media are generally of lower mole...

Destruction of Recombinant Tissue Plasminogen Activator (rtPA) -Loaded Echogenic Liposomes under Dual Frequency Sonication

Background:Echogenic liposomes (ELIPs) encapsulate drugs and gas bubbles within lipid vesicles. The destruction of ELIPs in response to MHz and kHz ultrasound waves has been studied previously. Applying ultrasound above a certain threshold causes encapsulated gas bubbles destruct rapidly by fragmentation or more slowly by acoustically driven diffusion. This study compares the d...

High molecular weight derivatives of human fibrinogen produced by plasmin. I. Physicochemical and immunological characterization.

Two high molecular weight derivatives of fibrinogen (Fragments X and Y) produced by plasmin digestion have been identified by comparative analyses of the digests in agar, agarose, and acrylamide electrophoresis, immunoelectrophoresis, Sephadex G-ZOO gel filtration, and analytical ultracentrifugation. These fragments were purified by salt precipitation and Sephadex G-200 gel fltration and their ...
